Ranunculus flabellaris Raf., the yellow water crowfoot, exhibitsstriking heterophylly between submerged and terrestrial leaves.Leaves produced under water are highly divided with numerousnarrow lobes and deep sinuses, whereas terrestrial leaves havefew broad lobes and shallow sinuses. When plants are submergedin a 25 µM solution of ABA, the typical transition fromterrestrial to submerged leaves is completely suppressed and,instead, terrestrial-like leaves are produced. Image analysistechniques show that, in addition to this modification of leafmorphology, leaves produced under ABA treatment possess surfaceand internal features characteristic of terrestrial leaf anatomy.This study provides evidence that the environmental factorsthat influence the morphological and anatomical expression ofheterophylly may act through endogenous ABA. Resting cysts and the excystment process in the freshwater ciliate Euplotes muscicola were studied by both light and scanning electron microscopy. Groups of distinctly crested resting cysts adhere to the substrate. Silver-stained preparations reveal surface conservation of dorsal kinetosomes and dorsal argyrome while ventral organelles are directed inward. Excystment involves the development of an expanding excystment vacuole concurrent with a localized thinning on the dorsal cyst wall surface. Cells exit through the pre-formed ostiole, mid-dorsal region first, initially by the force of cytoplasmic streaming, but later aided by cirral movement. Newly emerged cells retain the excystment vacuole and show no dorsal ridging. As the cell expels its excystment vacuole and partially unfolds, normal trophont morphology is re-established. Both cyst structure and cyst typology have implications for hypotrich taxonomy.  相似文献

The effects of para-fluorophenylalanine (p-FPA) and azetidine-2-carboxylicacid (AZ) on acid phosphatase and peroxidase activity, and onL-leucine incorporation, in root segments of Triticum aestivumL. cv. Fenman and Vigna radial a (L.) Wilczek, were studied.Incubation with 50 mmol–3 AZ significantly reduced phosphataseand peroxidase activities in wheat roots, but with 20 mol m–3p-FPA, only the peroxidase activity was reduced. In mung beanroots, phosphatase activity was inhibited by both AZ and p-FPA.Effects of the ortho- and meta- isomers of FPA on wheat rootphosphatase and peroxidase, and on mung bean phosphatase, werealso examined. Leucine uptake and incorporation were not inhibitedby 5 h pre-incubation with either p-FPA or AZ, but were inhibitedafter 24 h of pre-incubation. The results support the view that,in the shorter term, the analogues inhibit enzyme activity bybecoming incorporated to produce non-functional protein and,in the longer term, metabolism is further affected by inhibitionof protein synthesis. When AVT (arginine vasotocin) was given neonatally during theperiod when the brain is undergoing sexual differentiation,increased growth of the reproductive organs was observed inadulthood. Injection of AVT after this neonatal period in immatureanimals led to diminished growth of the accessory organs andin some cases the gonads themselves. The hypertrophic responseof the in situ ovary in adult mice following unilateral ovariectomy(UO) was inhibited in a dose-related manner by a single intraperitonealinjection of freshly prepared AVT. Much less AVT was requiredfor this response when injected into the third ventricle. Afterintraperitoneal injection, arginine vasopressin (AVP), lysinevasopressin (LVP), and 4-leucine vasotocin (4-leu-AVT) alsoinhibited compensatory ovarian hypertrophy whereas oxytocindid not. The commonality in die structure of these antigonadotrophicpeptides include a closed ring and a basic amino acid in position8. After opening the disulfide bond of these nonapeptides withmercaptoethanol, a single injection of the reduced AVT, AVP,LVP, or 4-leu-AVT into UO mice causes exaggerated hypertrophyof the remaining ovary. When added with leuteinizing hormone-releasinghormone (LRH) to culture medium containing hemipituitaries fromcastrated estrogen-progesterone primed female rats, AVT significantlyincreased the release of radioimmunoassayable LH above thatdue to LRH alone. Extracts from developing seeds, seedings, and different organsof mature plants of Sinapis alba L. were tested for the presenceof myrosinase. Three different patterns of isoenzymes were distinguishedby isoelectric focusing. One was found in developing seeds andseedlings, a second in mature leaves, stems, flowers, and podsand a third in roots. Sections of all the parts tested were examined for the presenceof myrosin cells by light microscopy. Developing seeds and seedlingsshowed a good correlation between the appearance of myrosinaseand the presence of myrosin cells. No myrosin cells were foundin root or flower parts although they showed myrosinase activity.  相似文献

The causes of cell wall necrosis in Blossom End Rot (BER) of tomatoes have been investigated by comparing enzyme activities in healthy and affected tissues in the cultivars Grenadier and Moneymaker. Changes in the mineral contents of the tomatoes were in accordance with those of calcium deficient tissues. The phenolic contents of the affected tissues were increased as also were caffeic and chlorogenic acids. Phenolase and peroxidase activities were higher in affected fruits white catalase activities were higher in healthy fruits. 14C-labelled leucine was incorporated into the proteins of the healthy fruits to the greatest extent but cell wall proteinbound hydroxyproline was greatest in affected fruits.  相似文献

Field trials confirmed that, whereas the Du Puits variety oflucerne was highly susceptible to Verticillium albo-atrum, ‘Gaetula’hybrids showed slight resistance and certain ‘Swedish’material was moderately resistant. Symptom-free ‘Gaetula’and Swedish plants were used to produce uninfected clonal materialby root propagation. Comparisons of the proteins of such plants with seed-grown DuPuits produced no evidence of a correlation between proteinpattern before inoculation and differences in susceptibilityand resistance. Disc electrophoretic comparisons of infected host material showedthat the pathogen gradually induced a change in the shoot proteinsof Du Puits whereas in the resistant host, after an initialmarked change, there was a development towards normality. Bythe terminal stages of wilting there was a noticeable fall inthe numbers of proteins in both roots and shoots of Du Puits,a finding confirmed both by double diffusion and immunoelectrophoresis.At the same time there was good evidence for a slight increasein protein bands in the resistant material after inoculation. After feeding tritium-labelled leucine to infected Du Puitsplants, a small protein component of fungal origin was detectedin the host. Labelled components from both healthy and infectedDu Puits plants were separated on Sephadex G.25 and compared.  相似文献
